CCAMF – Laboratorul de Imunologie a fost construit și echipat în 2015, datorită proiectului POS CCE O 2.2.1-2013-1: CENTRU AVANSAT DE CERCETARI MEDICALE SI FARMACEUTICE / CCAMF – Nr. 1837/48736. Laboratorul include dispozitive pentru separarea celulelor / modificare genetică / cultură / analiză: citometru cu flux cu 4 laser (sortator de celule BD FACSAria™), sisteme de separare celulară, ultracentrifugă, sistem de electroporare, microscop confocal (Leica TCS SP8), microscoape inversate (seria Leica), contoare celulare și, de asemenea ,analizatoare bazate pe tehnici precum: – tehnologia xMAP (FlexMap 3D) și Array (pe biocipuri), ELISA, CLIA, nefelometrie, turbidemetrie, spectrofotometrie, coagulometrie și aggregometrie – linie completă de electroforeză / bloting, sistem electroforetic automat pentru separarea și identificarea proteinelor (BioRad).

Personalul laboratorului este dedicat activităților de cercetare și pregătit să ofere asistență tehnică și să contribuie cu resurse intelectuale la diverse proiecte de cercetare.

Tehnologiile xMAP oferă detectarea simultană a peste 500 de markeri în suspensie sau pe suport solid, însoțite de tehnici ELISA, CLIA și nephelometrice. Aceste tehnici permit determinarea simultană a aspectelor calitative și cantitative ale unui număr mare de biomarkeri, semnale intracelulare, citokine, chemokine, factori de creștere, markeri metabolici, receptori nucleari și factori de transcripție.

Citometrul de flux identifică mai multe populații celulare într-un singur eșantion. Aceasta maximizează informațiile obținute din cantități limitate de probă și crește rentabilitatea testelor, deoarece poate reuni diverse informații (egale cu numărul de canale de detecție) despre sute de mii de celule într-o suspendare. Aplicații pentru analiza celulelor și elemente importante:

Separarea celulară – până la 4 linii celulare pure, concomitent

Imunofenotipizare (markeri de suprafață și marcatori intracelulari, măsurarea celulelor rare circulante)

Funcții celulare, apoptoză/viabilitatea celulară, analiza ciclului celular, proliferarea celulară

BD FACSDiva software te ajută să treci de la analiză la sortare. Sortatorul este echipat cu lasere de 405nm, 488nm, 561nm și 633nm și este capabil de detectare simultană de până la 9 culori. Populațiile pot fi sortate în până la 4 tuburi, care pot fi de asemenea controlate termic prin utilizarea unui bloc de sortare încălzit cu baie de apă disponibil. Populațiile sau celulele individuale pot fi, de asemenea, sortate direct pe lamele sau pe plăci standard sau personalizate.

Microscopia confocală este o tehnică de imagistică optică pentru creșterea rezoluției și contrastului optic, eliminând lumina neclară. Permite reconstrucția structurilor tridimensionale din imaginile obținute. Aplicații: scanare confocală (imagini cu laser) ale specimenelor vii și fixe marcate cu fluorescență, precum și pentru măsurători cantitative în domeniul științelor vieții. Puncte forte: Microscopia cu scanare cu laser confocal (CLSM) este una dintre serii de metode de generare a tăieturilor din probe microscopice prin intermediul opticii. Până la patru fluorofori plus o setare de câmp luminos pot fi imagistice în paralel sau secvențial. Există patru detectoare interactive de emisii, oferind utilizatorului flexibilitate în alegerea lungimii de undă de detectat. Beneficiul imagisticii confocale este un contrast crescut prin eliminarea neclaritatilor.
